G6PD deficiency is restricted to certain populations or segments of populations; therefore it may be that not every person requiring primaquine will be tested for their G6PD status, only those considered high risk for potential deficiency.

    Drug names in RED are of high risk to all deficiency variants. Names in GREEN are of low risk. Screening for G6PD deficiency thus seemed pointless and was not routinely done by the US Army until about 2006. During the 1960s, primaquine therapy against relapse was routinely administered without knowing G6PD status of the patient. Providers were warned to be alert to the risk of haemolytic anaemia and to stop dosing with onset of signs. There is also some evidence that quinine can cause haemolysis in patients with G6PD deficiency; such patients may also have increased blood concentrations of mefloquine when taken concurrently with primaquine.
