    The Addition of Chloroquine to Chemoradiation for Glioblastoma, Study Purpose Glioblastomas GBM are the most common type of primary brain tumors with an annual incidence of approximately 500 patients in the Netherlands. In addition to the immunotherapy trials specifically for patients with brain tumors, patients with glioblastoma often are eligible to receive new immunotherapy agents in clinical trials that are open to patients with any type of solid tumor through the Department of Investigational Cancer Therapeutics. Three cohorts of 3 patients will receive chloroquine in escalating doses 3 dose levels 200 mg up to 600 mg daily during standard treatment radiotherapy and temozolomide for newly diagnosed GBM. Extra patients can be added to a cohort in case of dose limiting toxicity, resulting in a maximum of 6 patients per dose level.

    The Centers for Disease Control and Prevention recommend against treatment of malaria with chloroquine alone due to more effective combinations. In areas where resistance is present, other antimalarials, such as mefloquine or atovaquone, may be used instead.
